I am currently trying to add wi-fi to my shed at the top of the garden which I am changing to a summer house. My question is can I run an Ethernet cable from my current BT Hub 2 up to the summer house and plug only into another BT Hub 2? Will this work and give me WiFi in the garden where it doesn't currently reach?

Yes, but unless you already have a spare Hub2, you would be better off buying a wireless access point (WAP)
If the shed is on the same mains consumer unit as the existing hub, you can simply use powerline adapters.
How does the WAP work? Would I still need to run an Ethernet up to the shed from my existing hub?
Yes, you are effectively considering using the second hub as a WAP so a simple WAP in the first place is an alternative if you are having to make a purchase.
However, powerline adapters obviate the need to run an Ethernet cable.
